Objective

The mission of the Joint Analysis Working Group is to explore, stimulate, facilitate and coordinate collaborative projects across the 4D Nucleome Network.

The Joint Analysis Working Group has defined two major goals during 4DN phase 2.

First, the working group defined a joint analysis project during phase I that focuses on benchmarking the diverse set of 4D Nucleome mapping technologies. The overarching goal of this project is to study the chromatin organization of a panel of common cell lines (H1-ESC, definitive endoderm derived from H1-ESCs and the fibroblast line HFFc6) using a diverse array of omics- and imaging-based technologies. Systematic comparisons of the different assays are being carried out to reveal the strengths and weaknesses of each method, to generate appropriate data standards, and to learn how different chromatin architectural features such as loops and TADs present themselves when examined using different methodologies. Results from this collaborative project are expected to greatly improve our understanding of nuclear organization and its dynamics in mammalian cells, and provide important guidelines for expanding such studies to additional cell types and biological contexts. The working group will publish the results of individual analyses in a series of papers, and a general synthesis in a “Flagship” publication.

Second, the working group aims to bring together representatives of all member groups of the 4D Nucleome Network to explore new collaborative projects. Once projects are defined, progress and coordination of the projects will be facilitated through bi-weekly meetings. The working group will closely interact with the 4DN data portal to enable data sharing between members and the 4D Nucleome Network in general.
